Well if the files are a type that can be compressed, like text files or uncompressed images/video (uncommon), then just use any archiver to zip them. I wouldn't expect to see too big of a difference though, since most space-consuming things now adays tend to be compressed already. Not sure about games though, that might help.

As a guess: MKVs will have varying results depending on what they contain (most things will already be compressed), and ISOs will probably yield decent results.
